Olanzapine(Zyprexa), Facts about Zyprexa,"ZYPREXA has been prescribed to over 6 million patients in 84 countries worldwide since its market entry in 1996. ZYPREXA is the first of the newer generation of antipsychotics (atypical antipsychotics) to be approved for long-term therapy in schizophrenia. As part of the newer class of atypical antipsychotics, ZYPREXA has benefits over the older, conventional antipsychotics in controlling both the positive (hallucinations, delusions) and negative (apathy, social withdrawal) symptoms. ZYPREXA is one of the most commonly prescribed treatments for schizophrenia. ZYPREXA is easy to use, with a recommended dosing schedule of once a day, with or without food. ZYPREXA is the first atypical antipsychotic approved for the short-term treatment of bipolar mania. In April, 2000, the Queen's Award for Enterprise in Innovation was granted by Britain's Queen Elizabeth II to Lilly United Kingdom for the discovery and successful implementation of ZYPREXA."


Olanzapine(Zyprexa), Uses, Health Center:"This medication acts to balance two nerve impulses and, by doing so, helps restore more normal thinking and mood. In general, this medication has fewer side effects than standard antipsychotic medications. Zyprexa also seems to help improve the negative symptoms of schizophrenia better than the other classes of antipsychotic medications."


Olanzapine, Indications, RXlist:"Olanzapine is indicated for the management of the manifestations of psychotic disorders. The antipsychotic efficacy of olanzapine was established in short-term (6-week) controlled trials of schizophrenic inpatients (see CLINICAL STUDIES ). The effectiveness of olanzapine in long-term use, that is, for more than 6 weeks, has not been systematically evaluated in controlled trials. Therefore, the physician who elects to use olanzapine for extended periods should periodically re-evaluate the long-term usefulness of the drug for the individual patient."


Olanzapine, Side Effects, MEDLINEplus:"Abdominal pain; awareness of heartbeat; decrease in sexual desire; double vision; fast heartbeat; increased appetite; increased cough; increased sensitivity of skin to sunlight; joint pain; low blood pressure; nausea; sore throat; stuttering; thirst; tightness of muscles; trouble in controlling urine; trouble in sleeping; vomiting; watering of mouth; weight loss."

Olanzapine, Drug class and Mechanism,"Olanzapine is a medication that is used to treat psychoses including schizophrenia . It is similar in chemical structure and in mechanism of action to clozapine (Clozaril). Like clozapine, olanzapine works by blocking receptors on the nerves of the brain for several neurotransmitters (chemicals that nerves use to communicate with each other). It binds to alpha-1, dopamine, histamine H-1, muscarinic, and serotonin type 2 (5-HT2) receptors. Olanzapine was approved by the FDA in 1996."


Olanzapine(Zyprexa), Olanzapine More Tolerable, As Effective As Clozaril For Treatment-Resistant Patients, October 30, 1998, Doctor's Guide:"Results of clinical studies of Eli Lilly and Co.s antipsychotic medication Zyprexa(R) (olanzapine) show that the drug is more tolerable and at least as effective as Clozaril (clozapine) in managing treatment-resistant patients."

Olanzapine, Effects of two atypical neuroleptics, olanzapine and risperidone, on the function of the urinary bladder and the external urethral sphincter in anesthetized rats, August, 2001, BioMed Central Pharmacology:"Olanzapine and risperidone significantly altered several voiding parameters and decreased the activity of the external urethral sphincter in the anesthetized rat. We propose that these effects are due to the central action of these drugs and not to peripheral effects. These findings may explain some of the clinical reports of urinary incontinence with risperidone and may predict similar occurrences with olanzapine therapy."


Olanzapine, Olanzapine Issued a Postive Opinion in Europe, June 20,"- Eli Lilly and Co said Thursday that Europe's Committee for Proprietary Medicinal Products issued a positive opinion for the marketing of Lilly's olanzapine, a drug the company hopes to market in the treatment of schizophrenia. The committee makes its recommendation to the European commission, which can issue a final, single license that is valid in all 15 European union countries. Lilly said it expects the approval process will be completed and early launches begun during the course of the year. The drug currently is not approved for marketing anywhere in the world."


Olanzapine, Interactions, Laurus Health:"Ingestion of alcohol may decrease blood levels of olanzapine by stimulating the liver to break down the drug. 2 Consequently, individuals who begin using alcohol while taking olanzapine may experience increased disease symptoms due to the reduced effectiveness of the drug. In addition, people who take antipsychotic agents such as olanzapine should avoid alcohol because it may intensify the effects of the drug on the nervous system and may cause low blood pressure."


Olanzapine, Pharmacodynamics, Mosby's DRUG Consult:"The mechanism of action of olanzapine, as with other antipsychotic drugs, is unknown. However, it has been proposed that this drug's antipsychotic activity is mediated through a combination of dopamine and serotonin type 2 (5HT2 ) antagonism. Antagonism at receptors other than dopamine and 5HT2 with similar receptor affinities may explain some of the other therapeutic and side effects of olanzapine. Olanzapine's antagonism of muscarinic M1-5 receptors may explain its anticholinergic effects. Olanzapine's antagonism of histamine H 1 receptors may explain the somnolence observed with this drug. Olanzapine's antagonism of adrenergic a1 receptors may explain the orthostatic hypotension observed with this drug."
